Scientific Description
The Czech Centre for Phenogenomics (CCP) is unique in combining genetic engineering capabilities, advanced phenotyping and imaging modalities, specific pathogen free (SPF) animal housing and husbandry, as well as cryopreservation and archiving, all in one central location – at BioCEV (www.biocev.eu/). This concentration of specialized infrastructure and expertise provides a unique and valuable resource for the medical research community.CCP through its membership in INFRAFRONTIER, is a partner in a collective global network that aims to comprehensively and systematically analyze the effect of loss of function gene mutations in mice. The goal is to produce a comprehensive ‘encyclopedia‘ of gene function, that will help identify causative factors of human diseases as well as novel targets for therapeutic intervention. Through the adoption of standardized procedures and pipelines, and the usage of quality control measures and cross-validation, our shared goal is to produce high-quality data that will not only function as a scientific reference catalog, but will also enable comprehensive meta-analyses that could uncover otherwise hidden disease relationships and functional interactions. By focusing expertise and resources and coordinating our efforts to avoid redundant work, we will help achieve these goals in a more cost-effective manner.

Embryo phenotyping

Services include imaging (microcomputed tomography and optical projection tomography), immunohistochemistry and in situ hybridization (section and whole-mount), alcian blue/alizarin red staining of cleared embryonic skeletons, late gestation vascular anatomy (india ink), beta-galactosidase staining.

Genetic engineering

Generation of custom genetically modified mouse and rat models by targeted mutagenesis and transgenesis (TALEN and Cas9/CRISPR, ES cell derivation, culture, electroporation and screening, pronuclear microinjection and blastocyst injections).

Animal housing

Mouse and rat husbandry and breeding (SPF facility, compliant with FELASA recommendations).

Adult phenotyping

Systematic standardized phenotyping in accordance with EMPReSS SOPs: assessment and measurement of behavior, metabolism, gross anatomy and body composition, hematology, clinical blood chemistry and urinalysis, immunology, cardiovascular and lung function.

Adult phenotyping

Equipment for echocardiography, electrocardiography, dual energy X-ray absoptiometry (DEXA), auditory brainstem response, fluorescence-activated cell sorting (FACS), indirect calorimetry, plethysmography, blood chemistry analysis, gait analysis, eye morphometry and behavioral analysis (open-field, water maze, elevated plus maze, Y-maze, startle reflex etc.).

Embryo phenotyping

Optical projection tomography, micro-computed tomography, dissecting microscopes etc.

Histopathology

Automated vacuum tissue processor, tissue embedding center, automated and rotary microtomes, cryostats, Ventana Symphony and Ventana BenchMark Special Stains.

Genetic engineering

micro-manipulating microscope systems, incubators, preparation and surgery microscopes, micropipette puller, microforge, laminar-flow benches, incubators, etc.

Animal housing

Breeding cages with racks (IVC and open), autoclaves, sterilization chambers, washing areas etc.

Archiving and distribution

controlled rate freezer, equipment for both ICSI and standard IVF, liquid nitrogen storage tanks with automated filling station etc.
